What is MK-677?
MK-677, also published as ibutamoren (MK-0677, L-163,191), is an orally active, non-peptide agonist of the ghrelin receptor (GHSR-1a). It was developed by Merck in the 1990s as a small-molecule alternative to injectable growth-hormone secretagogue peptides, and its research interest comes from its ability to stimulate pulsatile growth hormone release and raise circulating IGF-1 in study subjects.
Ibutamoren was taken into clinical studies on growth-hormone deficiency, age-related frailty and recovery after hip fracture, but it has never been approved as a medicine. Researchers usually compare it with peptide secretagogues because it shares their receptor target while being a non-peptide spiro-piperidine molecule, which is why it is stable enough to be supplied as a liquid solution or as a dry powder.
